I say yes. After Clijsters coming back and defeating Serena, here is Justine in only her second tournament back with even more on the line because it was a major final. I think this was the most significant victory for Serena coming off the US Open debacle. She made a pretty big statement here and shut up a lot of the doubters.
2007 is a very close second. As a matter of fact I think they are almost equally important for different reasons.

And, like you say, all for entirely different reasons.
AO03 - The significance of this win goes without saying. Serena Slam Completed, place in the history books etched
AO07 - Who gave her a chance in the media? Not many. She showed that she was and is the most determined and talented female tennis player on the planet (all due respect to the other ladies, esp. Venus and Justine)by some margin!
AO10 - This match was probably more important to 'Real' women's tennis fans, who have followed the tour avidly for years and know these two players to be, along with Venus an equal second with Henin, the best players of their generation. Serena losing a second slam in a row to a returning Belgian, would have just infuriated her I sense, plus, she shut up the media about her Little US Open incident
(I hope because frankly I'm bored of it).
So yes, it's hard for me to put this down as anything other than no.1, Victory at the Aussie was sweet against Dinara, for so many reasons, but knowing she did it against a player noone can undermine, makes it all the sweeter. Her bitterest rival, dispatched (albeit with a fight
) on the biggest stage in tennis! She must feel good about this one.
